Agro-industrial residues and starch for growth and co-production of polyhydroxyalkanoate copolymer and α-amylase by Bacillus sp. CFR-67 BJM

Polyhydroxyalkanoates (PHA) and α-amylase (α-1,4 glucan-4-glucanohydrolase, E.C. 3.2.1.1) were co-produced by Bacillus sp. CFR-67 using unhydrolysed corn starch as a substrate. Bacterial growth and polymer production were enhanced with the supplementation of hydrolysates of wheat bran (WBH) or rice bran (RBH) individually or in combination (5-20 g L-1, based on weight of soluble substrates-SS). In batch cultivation, a mixture of WBH and RBH (1:1, 10 g L-1 of SS) along with ammonium acetate (1.75 g L-1) and corn starch (30 g L-1) produced maximum quantity of biomass (10 g L-1) and PHA (5.9 g L-1). Assessing in vitro solubilization potential of different zinc solubilizing bacterial (zsb) isolates BJM

Zinc solubilizing ability of Bacillus sp. and Pseudomonas sp. was assessed using zinc oxide, zinc sulphide (sphalerite) and zinc carbonate in both plate and broth assays. ZSB-O-1 (Bacillus sp.) showed highest dissolution in the zinc sulphide (Sphalerite ore), with 2.80 cm of dissolution zone and 14.50 cm² of area in the plate assay and 13.60 mg kg-1 of zinc in the broth assay on the 15th day after inoculation. The ZSB-S-2 (Pseudomonas sp.) showed more solubilizing ability in the zinc oxide, with 3.30 cm clearing zone and 20.43 cm² area in the plate assay and 16.40 mg kg-1 of zinc in the broth assay over the same inoculation period. Objetivou-se selecionar e caracterizar bactérias do gênero Bacillus em rizosfera de Brachiaria brizantha como promotoras de crescimento de plantas e correlacionar os diferentes critérios de seleção das bactérias utilizados em laboratório e casa-de-vegetação. Após coleta de amostras de plantas em três municípios da região oeste de São Paulo, foram realizados o isolamento de bactérias da rizosfera e posterior caracterização fenotípica quanto ao potencial de colonização de raízes de braquiária, antagonismo a fungos fitopatogênicos, produção de auxinas, fosfatases e proteases, além da caracterização genotípica utilizando a técnica de PCR-RAPD. O controle químico é o método mais usado no manejo de doenças em grandes culturas como soja e trigo. No entanto, a alguns anos o controle biológico tem ganhado destaque. Assim, avaliou-se o antagonismo das bactérias Pseudomonas fluorencens, Pantoea aglomerans e Bacillus sp. sobre os fitopatógenos Corynespora cassiicola e Drechslera tritici-repentis, isolados previamente de folhas de soja e trigo, respectivamente. Non-symbiotic diazotrophic bacteria are amongst the most important functional groups of soil-dwelling microorganisms. These bacteria contribute to plant growth predominantly through biological N2 fixation. Here, we evaluated the density and diversity of non-symbiotic diazotrophic bacteria in soils taken from diverse land use systems (LUS) in Amazonia using nitrogen-free media. Abstract Various studies have been undertaken to evaluate the effect of polymer-based encapsulation on performance of plant growth promoting microorganism. However there is no systematic analysis about the performance of these microorganisms when encapsulated in a polymer-based matrix. Relevant published papers were retrieved by conducting searches in Google Scholar, Only Library, Science Direct, Springer Journal, Taylor & Francis and Wiley Online Library. Endophytic bacterial diversity in banana 'Prata Anã' (Musa spp.) roots Genet. Mol. Biol.

The genetic diversity of endophytic bacteria in banana 'Prata Anã' roots was characterized. Two hundred and one endophytic bacteria were isolated, 151 of which were classified as Gram-positive and 50 as Gram-negative. No hypersensitivity response was observed in any of the isolates. The rep-PCR technique generated different molecular profiles for each primer set (REP, ERIC and BOX). Fifty readable loci were obtained and all of the fragments were polymorphic. Amplified ribosomal DNA restriction analysis (ARDRA) of the isolates based on cleavage with four restriction enzymes yielded 45 polymorphic bands and no monomorphic bands. Highly thermostable and alkaline α-amylase from a halotolerant-alkaliphilic Bacillus sp. AB68 BJM

An alkaliphilic and highly thermostable α-amylase producing Bacillus sp. was isolated from Van soda lake. Enzyme synthesis occurred at temperatures between 25ºC and 40ºC. Analysis of the enzyme by SDS-PAGE revealed a single band which was estimated to be 66 kDa. The enzyme was active in a broad temperature range, between 20ºC and 90ºC, with an optimum at 50ºC; and maximum activity was at pH 10.5. The enzyme was almost completely stable up to 80ºC with a remaining activity over 90% after 30 min pre-incubation. Thermostability was not increased in the presence of Ca2+. An average of 75% and 60ºC of remaining activity was observed when the enzyme was incubated between pH 5 and 9 for 1 h and for 2 h, respectively. The activity of the enzyme was inhibited by...

ABSTRACT Forty isolates of endophytic bacteria isolated from banana tree roots were assessed as to their capacity to solubilize phosphate in a solid culture medium supplemented with different inorganic and one organic source of phosphorus. The amount of phosphorus (P) in each liquid medium was quantified, and an indirect assessment of acid phosphatase activity was performed. All assays had a fully randomized design, with three repetitions. Production and properties of alpha-amylase from thermophilic Bacillus sp. BJM

alpha-amylase (1,4-alpha-D-glucan glucanohydrolase, EC 3.2.1.1) production by thermophilic Bacillus sp strain SMIA-2 cultivated in liquid media containing soluble starch reached a maximum at 48h, with levels of 57U/mL. Studies on the a-amylase characterization revealed that the optimum temperature for activity was 70ºC. The enzyme was stable for 2h at 50ºC, while at 60ºC, 70ºC and 90ºC, 4%, 13% and 38% of the original activities were lost, respectively. The optimum pH of the enzyme was 7.5. After incubation of crude enzyme solution for 24h at pH 7.5, a decrease of about 5% of its original activity was observed. Production and properties of an extracellular protease from thermophilic Bacillus sp BJM

Protease production by thermophilic Bacillus sp strain SMIA-2 cultivated in liquid cultures containing trisodium citrate reached a maximum in 9h, with levels of 1.93U/mg protein. The microorganism utilized several carbon sources for the production of protease. Starch was the best substrate, followed by trisodium citrate, citric acid and sucrose. Among the various organic and inorganic nitrogen sources, ammonium nitrate was found to be the best. Studies on the protease characterization revealed that the optimum temperature of this enzyme was 60ºC. Studies on the stability of protease from Bacillus sp. and its compatibility with commercial detergent BJM

Enzymes, and particularly proteases, have become an important and indispensable part of industrial processes such as laundry detergents, pharmaceuticals and food products. Detergents such as Tide®, Ariel® and Biz® contain proteolytic enzymes, most of them produced by members of the genus Bacillus. This paper describes the compatibility of protease produced by the thermophilic Bacillus sp, with commercial laundry detergent. Stability studies indicated that this enzyme retained about 95% and 74% of its maximum activity after 1h at 60ºC in the presence of glycine in combination with MnSO4 and CaCl2, respectively. No inhibitory effect was observed at 1.0-5.0 mM of EDTA. Triton X-100 inhibited the enzyme in all the concentrations tested. The enzyme was unstable...
